Lenacapavir (LEN) is a long-acting first-in-class inhibitor of human immunodeficiency virus-1 (HIV-1) capsid protein that is in clinical development for the treatment and prevention of HIV-1 infection.¹ It can be taken orally (daily or weekly) or by subcutaneous injection (every 6 months).¹ LEN exerts its effects by inhibiting nuclear transport, capsid assembly, as well as virus assembly and release.¹

The CALIBRATE study was designed to study LEN’s effects in HIV patients who were antiretroviral (ARV) naïve patients.¹

In the study, subjects were randomized to 1 of 4 open-label once-daily groups (2:2:2:1), with differences in the 3 maintenance combinations at week 28 [once every 6 months (Q6M) LEN injections + emtricitabine (F) and tenofovir alafenamide (TAF), Q6M LEN injections + bictegravir (B) or oral LEN+F/TAF], compared with a control group of B/F/TAF (n=25); groups 1, 2, 3, and 4, respectively.¹
